Immunocytochemical Detection of MCF-7 Human Breast Dreast Cancer Cells Using Alkaline Phosphatase Antialkaline Phosphatase Method

Abstract
For a recent several years, there were several reports that occult micrometastasis could be detected in the bone marrow of breast cancer patients using a epithelial specific cytokeratin antibody in a cytochemistry assay. Immunofluorescence method
has
the disadvantage that morphological differentiation between tumor cells and cells of cross reactivity are not clear.
But immunocytochemical method carries the davantage that morphological diagnosis can be achieved and the slide can be mounted permanently. Among immunoenyme assay, alkaline phosphatase-antialkaline phosphatase(APAAP) method is more senistive than
others.
We used APAAP method with utilization of AE1/AE3 anticytokeration monoclonal antibody to detect the MCF-7 cells. We used MCF-7 cells and the mixture of MCF-7 cells and normal bone marrow cell as a test specimen. We also checked the morphology of
the
stained cell and checked the creoss reactivtiy of the normal bone marrow cell.
The detected cell was larger size and higher nuclear-cytoplasmic ratio than the normal bone marrow cell.
Almost all of the MCF-7 cells was identified by light microscopy since thelr cytoplasm was strongly stained by Fast Red Tr. Since there was no specific cross reactivity, there was no difficulty in differentiation of the MCF 7 cells from normal
bone
marrow cells.
